چکیده

Non-nucleoside reverse transcriptase inhibitors (NNRTIs) have, in addition to the nucleoside reverse transcriptase inhibitors (NRTIs) and protease inhibitors (PIs), gained a definitive place in the treatment of HIV-1 infections. The present work deals with computational ligand docking methodology, AutoDock 4.0, based on Lamarckian genetic algorithm for virtual screens of a compound database of 36 entries (tri-substituted 1,2,4-triazoles) for novel and selective inhibitors of the enzyme Reverse transcriptase (PDB entry;1RT2), a potential anti-AIDs drug target. Considering free energy of binding and inhibition constant (KI) as a criterion of evaluation, a total of 34 compounds were predicted to be potential inhibitors of reverse transcriptase and 14 compounds displayed greater binding affinities than Nevirapine, a well-known reverse transcriptase inhibitor. Compound AM31, 2-{[4-amino-5(2hydroxyphenyl)-4H-1, 2, 4-triazol-3-yl]-thio}-N-(4-nitrophenyl)acetamide; compound AM33, 2-{[4-amino-5-(2hydroxyphenyl)-4H-1,2,4-triazol-3-yl]-thio}-N-(4methoxyphenyl) acetamide; and compound AM34, 2-{[4-amino5-(2-hydroxyphenyl)4H-1,2,4-triazol-3-yl]thio}-N-(4-ethoxyphenyl)acetamide were considered to be the most potent reverse transcriptase inhibitors. Putative interactions between reverse transcriptase and inhibitors were identified by inspection of docking-predicted poses. Most of the compounds under study have shown significant binding energy as well as interaction in nanomolar range, thus suggesting the effectiveness of Autodock as an effective desktop molecular modelling tool. Attempts at discovering broad spectrum antiviral agents are presented herein.

Synthesis and molecular docking of novel N-((2-chloroquinolin-3-yl) methylene)-4-methylbenzenamine derivatives as anti-HIV-1 reverse transcriptase inhibitors

In this research work, a proficient method has been developed for the preparation of novel N-((2-chloroquinolin-3-yl) methylene)-4-methylbenzenamine derivatives from 2-chloroquinoline-3-carbaldehyde derivatives and p-toluidine in ethanol as solvent and using catalytic amount of acetic acid under reflux conditions to obtain desired products in good yields. The identification of all the synthesiz...

Novel acyclic nucleoside analogues as inhibitors of HIV-1 RT

Background Nucleoside reverse transcriptase inhibitors (NRTIs) were the first drugs introduced for treatment of human immunodeficiency virus-1 (HIV-1) infection. These NRTIs may be cyclic or acyclic analogs of natural nucleosides. Both these analogs interact at active site on HIV-RT and compete with indigenous nucleosides/ nucleotides, and thus, divert enzyme activity in manmade direction. All ...

Mitochondrial toxicity of nucleoside analogue reverse transcriptase inhibitors: a looming obstacle for long-term antiretroviral therapy?

Nucleoside reverse transcriptase inhibitors suppress HIV replication by blocking reverse transcriptase, an RNA-dependent DNA polymerase. These drugs can also affect cellular and mitochondrial DNA polymerases. Mitochondrial DNA polymerase gamma is particularly sensitive to nucleoside reverse transcriptase inhibitors, and the majority of adverse effects caused by nucleoside reverse transcriptase ...

Human immunodeficiency virus reverse transcriptase. General properties and its interactions with nucleoside triphosphate analogs.

Using affinity purified human immunodeficiency virus (HIV) reverse transcriptase the reaction assay conditions were determined. The optimum incorporation of dTMP into the (rA)n(dT)10 template with HIV reverse transcriptase required 6 mM MgCl2 and 80 mM KCl. The template specificity of HIV reverse transcriptase is quite different from those of the human gamma-polymerase-associated reverse transc...

Molecular Docking Studies of Novel Pyrazole Analogs as Possible Hiv-1 Rt Inhibitors

Human immunodeficiency virus (HIV) is the causative agent of the acquired immunodeficiency syndrome (AIDS) that has been affected 34 million people worldwide. Though an unprecedented growth has been taken place in the development of anti-HIV drugs during the last two decades, there is still no cure for this deadly disease. Cross resistance to anti HIV drugs is also a major concern in the develo...
